So I got the game today and first thing I did was put myself in, my parents and my two siblings (brother and sister), theyre the only two "children" (being 17 and 12) and I set both their relationships to me as sibling then the mii of my sister fell in love with my brother.
You can't assign family relationships to your non main mii in Tomodachi Collection either. It becomes weird when miis you don't want getting involved because they are related in real life or...
I can confirm that half siblings go under the siblings tab. Half siblings talk about each other just like siblings do and even mention a half sibling in connection with parents. And, just like full siblings, boy and girl half siblings have friendship ratings rather than romance ratings.
